Dosage
Adults: 10 mg up to 3 times daily, before meals. Maximum 7-day course at over-the-counter strength.
Side effects
Dry mouth, headache, abdominal cramps. Rare but serious: QT prolongation, especially at higher doses or in older adults.
Contraindications
Known QT prolongation, severe hepatic impairment, concomitant CYP3A4 inhibitors (ketoconazole, erythromycin).
Description
Domperidone blocks peripheral dopamine D2 receptors in the upper GI tract, accelerating gastric emptying and suppressing the vomiting reflex without crossing the blood-brain barrier.
